Lab-on-a-Chip for sample preparation and isolation of nucleic acids

In cooperation with the Institute for Military Science (Wehrwissenschaftliches Institut) Munster a chip-based microfluidic system was developed for the fully automated sample preparation and nucleic acid isolation from aqueous air samples. Hereby a commercially available spintube-based DNA preparation kit (Fa. Molzym) is transferred into a microfluidc system. The single functionalities of the kit (concentration of bacteria, lysis, DNA purification) were first established individually on single manual modules which in the next step were then automated and finally combined into an integrated chip. In addition, IMM developed a device capable to perform the preparation fully automatic. Accordingly, a suitable heating concept, valves and pumps were identified or developed to fulfill the demands of the fluidics and biology on chip.
